    This study aimed to examine the cellular and molecular events that occur in rhesus monkey testes after scrotal hyperthermia. Eight male adult rhesus monkeys were subjected to scrotal hyperthermia at 43°C for 30 min daily for 6 consecutive days. Sperm concentration, reproductive hormones, and testis histology were examined before hyperthermia (day 0), and at 8, 15, 30, 45, 60, 75, and 90 days after the initiation of hyperthermia. iTRAQ-based proteomic analysis was conducted on testicular tissues collected on days 0, 8, and 60 to identify differentially expressed proteins at the early and recovery stages of testicular damage. The sperm concentration was significantly decreased at days 30 and 45 after treatment (p < 0.01) and recovered to baseline at day 60. When compared with day 0, 101 and 24 differentially expressed proteins were identified at days 8 and 60 after heat treatment, respectively. The molecular functions of the differentially expressed proteins at day 8 were mainly nucleic acid binding, unfolded protein binding, nucleotide binding, and nucleoside phosphate binding. Spliceosome was enriched as the most significant pathway at day 8. CIRBP, PSIP1, Sam68, and Decorin were validated and found to be consistent with the proteomic data, indicating the reliability of the proteomic profiles identified in this study. In summary, we suggest that the proteins identified in this study may play important roles in heat-induced spermatogenic impairment. Some of these proteins, such as CIRBP, PSIP1, Sam68, and Decorin, may be early molecular targets responsible for spermatogenesis suppression induced by heat treatment.

    Levosimendan, a known calcium sensitizer with positive inotropic and vasodilating properties, might also be cardioprotective during ischemia-reperfusion (I/R) insult. Its effects on calcium homeostasis and apoptosis in I/R injury remain unclear. Na(+)/Ca(2+) exchanger (NCX) is a critical mediator of calcium homeostasis in cardiomyocytes, with reverse-mode NCX activity responsible for intracellular calcium overload and apoptosis of cardiomyocytes during I/R. We probed effects and underlying mechanisms of levosimendan on apoptosis and NCX activity in cultured human cardiomyocyte progenitor cells (CPC)-derived cardiomyocytes undergoing anoxia-reoxygenation (A/R), simulating I/R in vivo. Administration of levosimendan decreased apoptosis of CPC-derived cardiomyocytes induced by A/R. The increase in reverse-mode NCX activity after A/R was curtailed by levosimendan, and NCX1 was translocated away from the cell membrane. Concomitantly, endoplasmic reticulum (ER) stress response induced by A/R was attenuated in CPC-derived cardiomycytes treated with NCX-targeted siRNA or levosimendan, with no synergistic effect between treatments. Results indicated levosimendan inhibited reverse-mode NCX activity to protect CPC-derived cardiomyocytes from A/R-induced ER stress and cell death.

    The concentrations of potentially toxic metals like lead (Pb), cadmium (Cd), and mercury (Hg) in healthy lactating mothers of Lahore city were estimated during 2020-2021 with the help of a flame atomic absorption spectrometer (FAAS). Seventy samples of breast milk were collected from two different age groups, namely, G-1 (25-30 years) and G-2 (31-40 years). The results showed that Cd contents were below the detection limit of the measuring instrument; however, the Pb and Hg contents were detected with great ease. The mean concentrations of Pb and Hg in the G-1 corresponding to the age between 25 and 30 years were observed to be 1.914 ± 0.493 μg/L and 10.432 ± 3.249 μg/L, respectively. For the G-2 with an age limit between 31 and 40 years, the concentrations of Pb and Hg were estimated to be 2.045 ± 0.502 μg/L and 11.527 ± 3.231 μg/L, respectively. The T-test analysis of concentrations of these toxic metals indicated a significant correlation between the content levels of Pb and Hg (p < 0.05). The observed values of toxic metals were significantly more frequent than those suggested by WHO (World Health Organization). An increase in the concentrations of Pb and Hg is directly associated with the location of the target population. As a result, it is turned out that the majority of the lactating woman with significantly high levels of Pb and Hg belongs to industrial areas of Lahore. To avoid such conditions, residential areas must be at larger distances, in addition, to strictly abiding by the environmental policies of the government.
