    The majority of nucleotide binding domain leucine rich repeats-containing (NLR) family members has yet to be functionally characterized. Of the described NLRs, most are considered to be proinflammatory and facilitate IL-1β production. However, a newly defined sub-group of NLRs that function as negative regulators of inflammation have been identified based on their abilities to attenuate NF-κB signaling. NLRP12 (Monarch-1) is a prototypical member of this sub-group that negatively regulates both canonical and noncanonical NF-κB signaling in biochemical assays and in colitis and colon cancer models. The role of NLRP12 in infectious diseases has not been extensively studied. Here, we characterized the innate immune response of Nlrp12(-/-) mice following airway exposure to LPS, Klebsiella pneumoniae and Mycobacterium tuberculosis. In response to E. coli LPS, Nlrp12(-/-) mice showed a slight decrease in IL-1β and increase in IL-6 production, but these levels were not statistically significant. During K. pneumoniae infection, we observed subtle differences in cytokine levels and significantly reduced numbers of monocytes and lymphocytes in Nlrp12(-/-) mice. However, the physiological relevance of these findings is unclear as no overt differences in the development of lung disease were observed in the Nlrp12(-/-) mice. Likewise, Nlrp12(-/-) mice demonstrated pathologies similar to those observed in the wild type mice following M. tuberculosis infection. Together, these data suggest that NLRP12 does not significantly contribute to the in vivo host innate immune response to LPS stimulation, Klebsiella pneumonia infection or Mycobacterium tuberculosis.

    Predictions of pore pressure and seepage discharge are the most important parameters in the design of earth dams and assessing their safety during the operational period as well. In this research, soft computing models namely multi-layer perceptron neural network (MLPNN), support vector machine (SVM), multivariate adaptive regression splines (MARS), genetic programming (GP), M5 algorithm, and group method of data handling (GMDH) were used to predict the piezometric head in the core and the seepage discharge through the body of earth dam. For this purpose, the data recorded by the absolute instrument during the last 94 months of Shahid Kazemi Bukan Dam were used. The results showed that all of the applied models had a permissible level of accuracy in the prediction of the piezometric heads. The average error indices for the models in the training phase were R2= 0.957 and RMSE= 0.806 and in the testing phase were equal to R2= 0.949 and RMSE= 0.932, respectively. The performances of all models except the M5 and MARS in predicting seepage discharge are nearly identical; however, the best is the MARS, and the weakest is the M5 algorithm.

    Polygonatum cyrtonema Hua (PC) and Bletilla striata (BS) are widely used and planted as tuber traditional Chinese medicinal materials (TCMMs). Cadmium (Cd) is one of the major causes of soil pollution and challenge to the quality and safety of TCMMs. Understanding the absorption and distribution of Cd is important for addressing the risks posed by its residues. As a result, the higher Cd translocation factor (TF) results in the lower Cd bioconcentration factor (BCF) in the PC tuber than that of BS attributed to a lower Cd concentration in the PC tuber, which guaranteed its safe utilization and edible safety under 1 mg·kg-1 Cd soil. Cd stress overall activated peroxidase (POD), catalase (CAT), and water-extractable polysaccharides in PC (PCP1) to exhibit better antioxidation, while the superoxide dismutase (SOD) in BS increased by approximately 206-277% to alleviate more severe oxidative damage. Particularly, Cd induced an increase in PCP1 higher than that of water-extractable polysaccharides of BS (BSP1) by approximately 335% to 1351%. PC exhibited effective strategies for alleviating Cd toxicity, including transferring Cd to nonmedicinal parts, increasing polysaccharides, and synergistically activating the enzymatic antioxidant system. This study expands the application for the safe utilization of low-Cd contaminated soil and provides novel insights for tuber TCMMs to alleviate Cd toxicity.
