    A 29-year-old male patient underwent an autologous bone marrow transplant. He was initially diagnosed with Hodgkin's lymphoma and treated with 12 cycles of chemotherapy. Three months later, he presented with intermittent fever and underwent an MRI scan and a brain biopsy. Eventually, he was diagnosed with progressive multifocal leukoencephalopathy. For effective treatment and a plan of action, such cases necessitate multidisciplinary board meetings with input from experts in surgery, pathology, cancer, and infectious diseases.

    Endoscopy has been widely used in diagnosing gastrointestinal mucosal lesions. However, there are still lack of objective endoscopic criteria. Linked color imaging (LCI) is newly developed endoscopic technique which enhances color contrast. Thus, we investigated the clinical application of LCI and further analyzed pixel brightness for RGB color model. All the lesions were observed by white light endoscopy (WLE), LCI and blue laser imaging (BLI). Matlab software was used to calculate pixel brightness for red (R), green (G) and blue color (B). Of the endoscopic images for lesions, LCI had significantly higher R compared with BLI but higher G compared with WLE (all P < 0.05). R/(G + B) was significantly different among 3 techniques and qualified as a composite LCI marker. Our correlation analysis of endoscopic diagnosis with pathology revealed that LCI was quite consistent with pathological diagnosis (P = 0.000) and the color could predict certain kinds of lesions. ROC curve demonstrated at the cutoff of R/(G+B) = 0.646, the area under curve was 0.646, and the sensitivity and specificity was 0.514 and 0.773. Taken together, LCI could improve efficiency and accuracy of diagnosing gastrointestinal mucosal lesions and benefit target biopsy. R/(G + B) based on pixel brightness may be introduced as a objective criterion for evaluating endoscopic images.

    Research on greywater reuse in water stressed areas is in full swing. However, the perception of greywater reuse is one of the least researched areas in West Africa, particularly in Sahelian countries. This study aimed to fills a significant gap in the existing literature, which has largely ignored the specific socio-demographic contexts of developing countries in the Sahelian regions. The study involved in-depth interviews with 240 rural households and the collection of 40 greywater samples in four locations for laboratory analysis. The survey focused on greywater management and household perceptions of greywater reuse in agriculture. The analyses focused on determining the physico-chemical and microbiological parameters of greywater collected from households. The results showed that over 80 % of households discharged greywater into the natural environment without prior treatment. The majority of respondents were aware that poor greywater management poses a health risk. The results also showed an association between locality, gender, education level and perceptions of poor grey water management. Respondents were willing to accept the reuse of greywater in agriculture, to consume irrigated vegetables and to install a greywater treatment system. The quality analysis showed that the greywater was not in compliance with the legal discharge limits. This study highlights that in order to promote sustainable greywater management practices within households, it is important to design effective greywater treatment systems that meet the needs of the target population. Awareness campaigns, education and training programmes on wastewater management could also be established.

    Postoperative delirium is a common postoperative complication of neurocognitive dysfunction, especially in elderly surgical patients. Postoperative delirium not only damages patients' recovery but also increases social costs. Therefore, its prevention and treatment has essential clinical and social significance. However, due to its intricate pathogenesis and limited pharmacological interventions, the effective prevention and treatment of postoperative delirium remains a thorny problem. As traditional acupuncture therapy has been shown to be an effective treatment in many neurological disorders, and in recent years, it has begun to be clinically used as an intervention for postoperative delirium. Although most clinical and animal studies confirm that multiple types of acupuncture interventions can alleviate or prevent postoperative delirium by relieving acute postoperative pain, reducing the consumption of anesthetics and analgesics, attenuating neuroinflammation and neuronal lesions, while more evidence-based medical evidence and clinical validation are needed for these encouraging effects.

    The study compares three variants of focus sensors designed for the optical topography measurement of rough surface specimens with submicron accuracy. We present a theoretical analysis of the focus sensor principles and the experimental measurements with a single point laser probe. A low coherent illumination beam was provided by a monochromatic laser source and a rotating diffuser, which reduced the speckles generated by the rough surface. The reflected beam was modulated by three specific optical elements (axicon, double wedge prism, four spherical lenses) realized by a spatial light modulator. A digital camera detected the output intensity patterns that were evaluated by the intensity centroid method. The results showed a good coincidence of the surface profiles obtained by the three sensor variants with the root-mean-square deviations below one micron. We discuss the results obtained for several specimens with various surface roughness and compare the differences between the three focus sensor variants.

    22q11 Deletion Syndrome (22q11DS) is associated with dysmorphology and a high prevalence of schizophrenia-like symptoms. Several genes located on chromosome 22q11 have been linked to schizophrenia. The deletion is thought to disrupt the expression of multiple genes involved in maturation and development of neurons and neuronal circuits, and neurotransmission. We investigated whole-genome gene expression of Peripheral Blood Mononuclear Cells (PBMC's) of 8 22q11DS patients and 8 age- and gender-matched controls, to (1) investigate the expression levels of 22q11 genes and (2) to investigate whether 22q11 genes participate in functional genetic networks relevant to schizophrenia. Functional relationships between genes differentially expressed in patients (as identified by Locally Adaptive Statistical procedure (LAP) or satisfying p<0.05 and fold-change >1.5) were investigated with the Ingenuity Pathways Analysis (IPA). 14 samples (7 patients, 7 controls) passed quality controls. LAP identified 29 deregulated genes. Pathway analysis showed 262 transcripts differentially expressed between patients and controls. Functional pathways most disturbed were cell death, cell morphology, cellular assembly and organization, and cell-to-cell signaling. In addition, 10 canonical pathways were identified, among which the signal pathways for Natural Killer-cells, neurotrophin/Trk, neuregulin, axonal guidance, and Huntington's disease. Our findings support the use of 22q11DS as a research model for schizophrenia. We identified decreased expression of several genes (among which COMT, Ufd1L, PCQAP, and GNB1L) previously linked to schizophrenia as well as involvement of signaling pathways relevant to schizophrenia, of which Neurotrophin/Trk and neuregulin signaling seems to be especially notable.

    The recently developed plasmonic and photonic metal-semiconductor nanolasers feature unique properties, such as ultra-small mode volume and footprint, high Purcell factor, and ultra-fast modulation. However, it is often difficult to recognize when the transition to lasing occurs, while the most important feature of laser radiation, i.e., coherence, is available only above the lasing threshold. Here we systematically study the second-order coherence properties of metal-semiconductor nanolasers at both low- and high-pump rates. We find the lasing threshold using a clear coherence definition and derive a simple expression for the threshold pump current (optical pump power), which can be applied to most thresholdless and non-thresholdless metal-semiconductor nanolasers.

    BACKGROUND: Anxiety is rising across the United States during the COVID-19 pandemic, and social distancing mandates preclude in-person mental health care. Greater perceived control over anxiety has predicted decreased anxiety pathology, including adaptive responses to uncontrollable stressors. Evidence suggests that no-therapist, single-session interventions can strengthen perceived control over emotions like anxiety; similar programs, if designed for the COVID-19 context, could hold substantial public health value.
    OBJECTIVE: Our registered report evaluated a no-therapist, single-session, online intervention targeting perceived control over anxiety in the COVID-19 context against a placebo intervention encouraging handwashing. We tested whether the intervention could (1) decrease generalized anxiety and increase perceived control over anxiety and (2) achieve this without decreasing social-distancing intentions.
    METHODS: We tested these questions using a between-subjects design in a weighted-probability sample of US adults recruited via a closed online platform (ie, Prolific). All outcomes were indexed via online self-report questionnaires.
    RESULTS: Of 522 randomized individuals, 500 (95.8%) completed the baseline survey and intervention. Intent-to-treat analyses using all randomized participants (N=522) found no support for therapeutic or iatrogenic effects; effects on generalized anxiety were d=-0.06 (95% CI -0.27 to 0.15; P=.48), effects on perceived control were d=0.04 (95% CI -0.08 to 0.16; P=.48), and effects on social-distancing intentions were d=-0.02 (95% CI -0.23 to 0.19; P=.83).
    CONCLUSIONS: Strengths of this study included a large, nationally representative sample and adherence to open science practices. Implications for scalable interventions, including the challenge of targeting perceived control over anxiety, are discussed.

    A transesterification reaction has been carried out over a transition metal modified hierarchical ETS-10 (METS-10) catalyst to synthesize glycerol carbonate (GC) from glycerol. The inherent Lewis basicity of ETS-10 favors oriented conversion of glycerol, and the hierarchical structure benefits exposure of more active sites, shortens the molecular diffusion path, suppresses the formation of coke in the micropores, and then enhances the catalytic reactivity and stability. Furthermore, the influence of transition metals (Fe, Co, Ni, Cu, Zn and Mn) on the basic sites of supports has been investigated in detail. It is found that basicity change of the catalyst depends on not only the cation size, nature and composition of the transition metal but also the zeolite structure and pore topology. Besides, the presence of a certain amount of Ni0 species from catalyst reduction has proved to play a critical role in strengthening the interaction of Lewis basic sites (TiO6 2-) with active glycerol hydroxyl groups. Finally, a 97.7% glycerol conversion and 97.1% GC yield have been obtained over Ni/METS-10, of which the high catalytic performance can be maintained after 8 runs.

    With the increasing concern regarding climate change, academics and practitioners are devoting attention to corporate environmental sustainability development. However, corporate environmental responsibility as an outcome of corporate governance (CG) practice is also constrained by national culture as an institutional factor, and research on the relationship between CG and environmental sustainability performance (ESP) with consideration for national culture remains scarce. Therefore, this study investigates the ESP data of Forbes' listed multinational corporations (MNCs) through content analysis and applies STATA software with stepwise regression models to empirically test the relationship between CG and MNCs' ESP and the moderating effects of national culture on this relationship. The results show that board independence and board size positively affect MNCs' ESP, and the relationship between board independence and MNCs' ESP is negatively moderated by masculinity and uncertainty avoidance. Our results emphasize the importance of CG in environmental decision-making by board management supervision enhancement and explain how national culture affects ESP because of its influence on CG. Our study explains the agency effect of board composition on MNCs' environmental sustainability development and the influence of national culture, which establishes a link between CG, ESP, and national culture. Moreover, policymakers and MNCs' suggestions for enhancing ESP through CG measures, while considering national culture, are also provided.

    Production of large quantities of organic waste all over the world poses major environmental and disposal problems. The present study was conducted to explore the deleterious effects of Parthenium hysterophorus and milk processing industry sludge on the health of earthworm, Eisenia fetida. Earthworms were allowed to grow in the mixture of cow dung:Parthenium hysterophorus (75:25) and cow dung:milk processing industry sludge (60:40) for 60 days. The biochemical markers viz. superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), and malondialdehyde (MDA) levels and histological changes in earthworm's intestine were assessed after 15, 30, 45, and 60 days of exposure. The results revealed increased MDA level, while SOD, CAT, and GPx activities showed variation in both treatments. Furthermore, histopathological changes revealed damage in the intestinal tissue in both treatments during all intervals. More severe effects were registered in P. hysterophorus treatment. Obtained results may contribute to the understanding of P. hysterophorus and milk processing industry sludge induced toxic effects on earthworms and to identify defense mechanism of Eisenia fetida.

    Tumor-derived exosomes are emerging noninvasive biomarker reservoirs that reflect biological information from their parental cells, especially specific markers, including proteins, DNA fragments and RNAs. Recently, analytical methods of tumor-derived exosomes have been increasing growth. However, developing a convenient signal amplification technique to improve the sensitivity of exosomes detection still remains a challenge. Herein, an ultrasensitive and specific exosomes diagnostic biochip is constructed and further applied to circulating tumor exosomes detection in serum. Using an exosomes diagnostic biochip, signal amplification is achieved by combining the advantages of quantum dots with the biomimetic periodic nanostructure of photonic crystals. Glypican-1 (GPC1), a membrane-anchored protein that is overexpressed in exosomes from pancreatic cancer, is detected using nanosized molecular beacons with high luminescence efficiency; then the signal is amplified through photonic crystals. Moreover, the method allows the quantitative analysis of various disease-specific surface proteins on exosomes. We believe that this exosomes diagnostic biochip is likely to have potential as an effective bioassay, which may be helpful for quantification of disease-specific exosomes in clinical use.

    P-Glycoprotein (Pgp)-medicated multidrug resistance (MDR) remains a formidable challenge to cancer therapy. As conventional approaches using small-molecule inhibitors failed in clinical development because of the lack of cancer specificity, we develop Pgp-targeted carbon nanotubes to achieve highly cancer-specific therapy through combining antibody-based cancer targeting and locoregional tumor ablation with photothermal therapy. Through a dense coating with phospholipid-poly(ethylene glycol), we have engineered multiwalled carbon nanotubes (MWCNTs) which show minimum nonspecific cell interactions and maximum intercellular diffusion. After chemically modifying with an anti-Pgp antibody, these MWCNTs showed highly Pgp-specific cellular uptake. Treatment of the targeted MWCNTs caused dramatic cytotoxicity in MDR cancer cells upon photoirradiation, whereas they did not cause any toxicity in the dark or phototoxicity toward normal cells that do not express Pgp. Because of excellent intratumor diffusion and Pgp-specific cellular uptake, the targeted MWCNTs produced strong phototoxicity in tumor spheroids of MDR cancer cells, a 3-D tumor model for studying tumor penetration and therapy. In conclusion, we have developed highly Pgp-specific MWCNTs that may provide an effective therapy for MDR cancers where other approaches have failed.

    H-Ras and N-Ras become activated both at the plasma membrane and in endomembrane structures such as the Golgi apparatus. This compartmentalized activation is relevant from a signaling standpoint, because effector molecules can become activated differently depending on the region of the cell where Ras proteins are activated. An unsolved question in this new regulatory mechanism is the understanding of how Ras proteins become activated in endomembranes. To approach this problem, we have studied the subcellular distribution and activities of a number of Ras guanosine nucleotide exchange factors. Our results indicate that Ras activation at the plasma membrane and endoplasmic reticulum is an unspecific process that can be achieved by most Ras activators. In contrast, GTP loading of Ras at the Golgi is only induced by members of the Ras guanosine nucleotide releasing protein family. In agreement with these observations, Ras guanosine nucleotide releasing proteins are the only Ras activators showing localization in the Golgi. These results indicate that the compartmentalized activation of effector pathways by Ras proteins depends not only on the specific localization of the GTPases but also in the availability of GDP/GTP exchange factors capable of activating Ras proteins in specific subcellular compartments.

    The binding of the B subunits of Escherichia coli heat-labile enterotoxin (LT) to epithelial cells lining the intestines is a critical step for the toxin to invade the host. This mechanism suggests that molecules which possess high affinity to the receptor binding site of the toxin would be good leads for the development of therapeutics against LT. The natural receptor for LT is the complex ganglioside GM1, which has galactose as its terminal sugar. A chemical library targeting a novel hydrophobic pocket in the receptor binding site of LT was constructed based on galactose derivatives and screened for high affinity to the receptor binding site of LT. This screening identified compounds that have 2-3 orders of magnitude higher affinity toward the receptor binding site of LT than the parent compound, galactose. The present findings will pave the way for developing simple and easily synthesizable molecules, instead of complex oligosaccharides, as drugs and/or prophylactics against LT-caused disease.

    Rare biallelic BLM gene mutations cause Bloom syndrome. Whether BLM heterozygous germline mutations (BLM+/-) cause human cancer remains unclear. We sequenced the germline DNA of 155 mesothelioma patients (33 familial and 122 sporadic). We found 2 deleterious germline BLM+/- mutations within 2 of 33 families with multiple cases of mesothelioma, one from Turkey (c.569_570del; p.R191Kfs*4) and one from the United States (c.968A>G; p.K323R). Some of the relatives who inherited these mutations developed mesothelioma, while none with nonmutated BLM were affected. Furthermore, among 122 patients with sporadic mesothelioma treated at the US National Cancer Institute, 5 carried pathogenic germline BLM+/- mutations. Therefore, 7 of 155 apparently unrelated mesothelioma patients carried BLM+/- mutations, significantly higher (P = 6.7E-10) than the expected frequency in a general, unrelated population from the gnomAD database, and 2 of 7 carried the same missense pathogenic mutation c.968A>G (P = 0.0017 given a 0.00039 allele frequency). Experiments in primary mesothelial cells from Blm+/- mice and in primary human mesothelial cells in which we silenced BLM revealed that reduced BLM levels promote genomic instability while protecting from cell death and promoted TNF-α release. Blm+/- mice injected intraperitoneally with asbestos had higher levels of proinflammatory M1 macrophages and of TNF-α, IL-1β, IL-3, IL-10, and IL-12 in the peritoneal lavage, findings linked to asbestos carcinogenesis. Blm+/- mice exposed to asbestos had a significantly shorter survival and higher incidence of mesothelioma compared to controls. We propose that germline BLM+/- mutations increase the susceptibility to asbestos carcinogenesis, enhancing the risk of developing mesothelioma.
