We developed an anaerobic digestate tablet using liquid dairy cattle manure derived from a small mesophilic anaerobic digester installed on a dairy farm. Anaerobic digestate tablets are intended for use in the fertilization of oligotrophic coastal seas to promote primary production. The purpose of this study was to evaluate (1) the dissolution behavior of nutrients from anaerobic digestate tablets and (2) the effect of the application of anaerobic digestate tablets on the growth of benthic microalgae using a culture experiment. Batch experiments were conducted to investigate the dissolution behavior of the nutrients. Cumulative amounts of dissolved inorganic nitrogen and phosphate in the anaerobic digestate tablet ranged from 110 to 28.9 μg g-1 after 28 days. The dissolved inorganic nitrogen in the anaerobic digestate tablet was mainly ammonium nitrogen and accounted for 92.4-96.9%, which is advantageous for the growth of microalgae. The growth curve of the benthic microalga Nitzchia longissima was monitored using f/2 medium added to the anaerobic digestate tablet. The growth of Nitzchia longissima was two orders of magnitude greater than that of the positive control. The enhanced growth of Nitzchia longissima by the anaerobic digestate tablet was considered a concomitant effect of moderate dissolution of ammonium nitrogen and high affinity for benthic microalgae. In conclusion, the anaerobic digestate tablets prepared in this study have the advantage of supplying nitrogen to benthic microalgae. This study proposes a new method for utilizing anaerobic digestates.

Detecting ZEN is crucial for assessing its potential health risks. This study developed a highly sensitive non-competitive magnetic phage anti-immunocomplex immunoassay (Nc-MPHAIA) for ZEN detection, utilizing the anti-ZEN single-chain variable fragment (ScFv) and anti-immunocomplex peptide (AIcP), both of which were screened using phage display technology. The method exhibited a limit of detection of 0.005 ng/mL, which was 112 times lower than that of the conventional competitive immunoassay using the same ScFv and the chemosynthetic antigen ZEN-OVA. Validation studies demonstrated excellent recovery rates in spiked corn samples (intra-assay: 84.04-109.95 %; inter-assay: 89.70-99.59 %). Additionally, the Nc-MPHAIA results for ZEN in natural samples were consistent with those obtained via high-performance liquid chromatography, which demonstrated good reliability of the established assay.

BLLB/c female mice conditioned with busulfan/cyclophosphamide (Bu-Cy) were transplanted with allogeneic male C57BL/6. Control group consisted of syngeneic transplanted Balb/c mice. In allogeneic settings, significant expansion and maturation of donor dendritic cells (DCs) were observed at day +3, while donor T-cells CD8+ were increased at day +5 (230%) compared to syngeneic HSCT. Highest levels of inflammatory cytokines IL-2, IFN-gamma, and TNF-alfa at day +5 matched T-cell activation. Concomitantly naïve T-cells gain effecr-memory phenotype and migrated from spleen to peripheral lymphoid organs. Thus, in the very early phase of GHVD following Bu-Cy conditioning donor, DCs play an important role in the activation of donor T cells. Subsequently, donor naïve T-cells gain effector-memory phenotype and initiate GVHD.
- ArticleYasuda K, Takahashi M, Mori N.PLoS One. 2015;10(11):e0142943.NatB is an N-terminal acetyltransferase consisting of a catalytic Nat5 subunit and an auxiliary Mdm20 subunit. In yeast, NatB acetylates N-terminal methionines of proteins during de novo protein synthesis and also regulates actin remodeling through N-terminal acetylation of tropomyosin (Trpm), which stabilizes the actin cytoskeleton by interacting with actin. However, in mammalian cells, the biological functions of the Mdm20 and Nat5 subunits are not well understood. In the present study, we show for the first time that Mdm20-knockdown (KD), but not Nat5-KD, in HEK293 and HeLa cells suppresses not only cell growth, but also cellular motility. Although stress fibers were formed in Mdm20-KD cells, and not in control or Nat5-KD cells, the localization of Trpm did not coincide with the formation of stress fibers in Mdm20-KD cells. Notably, knockdown of Mdm20 reduced the expression of Rictor, an mTORC2 complex component, through post-translational regulation. Additionally, PKCαS657 phosphorylation, which regulates the organization of the actin cytoskeleton, was also reduced in Mdm20-KD cells. Our data also suggest that FoxO1 phosphorylation is regulated by the Mdm20-mTORC2-Akt pathway in response to serum starvation and insulin stimulation. Taken together, the present findings suggest that Mdm20 acts as a novel regulator of Rictor, thereby controlling mTORC2 activity, and leading to the activation of PKCαS657 and FoxO1.
- ArticleFan X, Bao Y, Mumtaz M, Huang J, Yu G.Sci Total Environ. 2021 Apr 01;763:142943.In this study, ultraviolet (UV)-activated persulfate under alkaline condition was developed as an alternative Total Oxidizable Precursor (TOP) assay to convert per- and polyfluoroalkyl substances (PFASs) precursors into detectable perfluoroalkyl acids (PFAAs). The conventional heat-based TOP assay takes 6 h at 85 °C, which is time consuming and may lose the volatile PFASs. Shorter time treatment would be beneficial to promote the analysis efficiency of the samples. We here report the UV-based TOP method as faster replacement of conventional heat-based TOP assay. The 6:2 fluorotelomer sulfonate (FTS), 8:2 FTS, and perfluorooctanesulfonamide (FOSA) achieved nearly complete conversion (with the molar yield of PFAAs of 97.2%-109.9%) at 60 min. This new UV-based TOP assay was then applied to 23 industry samples, and the results are comparable with the heat-based TOP assay. The perfluoroalkyl carboxylic acids (PFCAs) concentrations of 23 samples before and after normal TOP assay were 0-4290 mg L-1 and 438-77,420 mg L-1, respectively. The PFCAs after 60 min UV-based TOP assay was 310-81,881 mg L-1. The QuotientMol of 21 samples were 0.71-1.28, the QuotientMol of other two samples were 1.45 and 1.68, this probably due to the loss of volatile precursors during heat-based TOP assay. The UV-based TOP assay is an efficient, reliable and faster alternative to the conventional heat-based TOP assay.
