Dysregulated circular RNAs (circRNAs) play a vital role in HCC. We aimed to study the role of circ_0003528 in HCC and its fundamental molecular mechanisms. HSA_circ_0003528 was identified through bioinformatics dataset analysis. The binding sites between mRNA and miRNA were predicted using online bioinformatics tools. The interaction between miR-212-3p and X-linked inhibitor of apoptosis protein (XIAP) or circ_0003528 was confirmed through the luciferase reporter assay. RT-qPCR and western blot assays were used to analyze the expression of all miRNAs/mRNAs and proteins. Cellular functions were evaluated using the MTT and TUNEL assays. A xenograft model was established to evaluate the function of circ_0003528 in vivo. Circ_0003528 was dramatically overexpressed in HepG2 and HUH7 cells. However, knockdown of circ_0003528 suppressed the aggressiveness of HCC cells and tumor growth both in vitro and in vivo. Furthermore, binding of miR-212-3p to circ_0003528 and XIAP was verified. Downregulation of miR-212-3p abrogated the effects of si-circ_0003528 on cell viability and apoptosis, and upregulation of XIAP antagonized the functions of the miR-212-3p mimic in HCC cells. circ_0003528 contributes to the development of HCC in vitro and in vivo via the miR-212-3p/XIAP axis. Hence, circ_0003528 knockdown may be a potential therapeutic strategy for HCC treatment.

The precursor compound, (NH4)2MoS4, was incorporated into H-MOPs. Heat treatment under argon resulted in the formation of hollow MoS2-carbon nanocomposites (MSC). According to microscopic analysis, the MoS2 in the MSC has a layered structure with an elongated interlayer distance. The MSC showed high reversible discharge capacities up to 802 mA h g(-1) after 30 cycles and excellent rate performance for lithium ion batteries. The promising electrochemical performance of the MSC is attributed to the very thin and disordered nature of MoS2 in the carbon skeleton. The role of chemical components of the MSC in the electrochemical process was suggested.

Propagules of varying magnitudes have been used to try and establish populations into novel locations, though the relationship between propagule size and species establishment remains speculative. Previous qualitative studies argue that site-level factors are of importance when determining where to release Chukar. We utilized machine learning ensembles to evaluate bioclimatic and topographic data from native and naturalized regions to produce predictive species distribution models (SDMs) and evaluate the relationship between establishment and site-level factors for the conterminous United States. Predictions were then compared to a distribution map based on recorded occurrences to determine model prediction performance. SDM predictions scored an average of 88% accuracy and suitability favored states where Chukars were successfully introduced and are present. Our study shows that the use of quantitative models in evaluating environmental variables and that site-level factors are strong indicators of habitat suitability and species establishment.

Data fusion using a Kalman filter and map matching are effective approaches to improve the performance of navigation system applications based on GPS/MEMS IMUs. The study considers the curve-to-curve matching algorithm after Kalman filtering to correct mismatch and eliminate redundancy. By applying data fusion and map matching, the study easily accomplished mapping of a GPS/INS trajectory onto the road network. The results demonstrate the effectiveness of the algorithms in controlling the INS drift error and indicate the potential of low-cost MEMS IMUs in navigation applications.

Clinical and self-reported periodontitis, along with sociodemographic data, were collected from 197 adults. Akaike information criterion models were developed to identify periodontitis, and nomograms developed based on its regression coefficients. The discriminatory capability was evaluated by receiver-operating characteristic curves. Decision curve analysis was performed. Smoking [OR 3.69 (95%CI 1.89, 7.21)], poor/fair self-rated oral health [OR 6.62 (95%CI 3.23, 13.56)], previous periodontal treatment [OR 9.47 (95%CI 4.02, 22.25)], and tooth loss [OR 4.96 (95%CI 2.47, 9.97)], determined higher probability of having "Moderate/Severe Periodontitis". Age [OR 1.08 (95%CI 1.05, 1.12)], low educational level [OR 1.65 (95%CI 1.34, 2.23)], poor/fair self-rated oral health [OR 3.57 (95%CI 1.82, 6.99)], and previous periodontal treatment [OR 6.66 (95%CI 2.83, 15.68)] determined higher probability for "Any Periodontitis". Both nomograms showed excellent discriminatory capability (AUC of 0.83 (95%CI 0.75, 0.91) and 0.81 (95% CI 0.74, 0.88), good calibration, and slight overestimation of high risk and underestimation of low risk. Hence, our nomograms could help identify periodontitis among adults in Denmark.

In the context of energy shortage and environmental pollution, it is very important to study lightweight and ensure car safety. In this paper, a lightweight fiber metal laminate (FML) with excellent cushioning and energy absorption characteristics for car body is proposed by studying the deformation and energy absorption mechanism of ordinary groove plate under flat pressure. The FML is made up of an M-shaped aluminum alloy sandwich plate and two carbon fiber reinforced composite outer panels. By means of experiment and finite element simulation, it is proved that this structure had good cushioning energy absorption effect. Under the same conditions, the peak force of the M-shaped corrugated core FML with 2.5 mm groove depth is 46.3% of that of the traditional trapezoidal corrugated-core FMLs. Besides, the energy absorption capacity of the M-shaped corrugated core FML with 2 mm groove depth is 1.62 times of that of the traditional trapezoidal corrugated-core FMLs, before the top plate of sandwich unit touches the bottom plate. And cushioning energy absorption mechanism of M-shaped corrugated core FML is further explained by simulation.

However, previous literature shows that a dilemma exists between polarization, endurance and retention. Since all these properties should be simultaneously high, overcoming this issue is of the highest relevance. Here, we demonstrate that high crystalline quality sub-5 nm Hf0.5Zr0.5O2 capacitors, integrated epitaxially with Si(001), present combined high polarization (2Pr of 27 μC cm-2 in the pristine state), endurance (2Pr > 6 μC cm-2 after 1011 cycles) and retention (2Pr > 12 μC cm-2 extrapolated at 10 years) using the same poling conditions (2.5 V). This achievement is demonstrated in films thinner than 5 nm, thus opening bright possibilities in ferroelectric tunnel junctions and other devices.

The reaction of [(PPh3)Au(L)](+) (L = Cl(-) or substituted N- heterocyclic pyridine) with the C-terminal (Cys3His) finger of HIVNCp7 shows evidence by mass spectrometry (ESI-MS) and (31)P NMR spectroscopy of a long-lived {(PPh3)Au}-S-peptide species resulting from displacement of the chloride or pyridine ligand by zinc-bound cysteine with concomitant displacement of Zn(2+). In contrast, reactions with the Cys2His2 finger-3 of the Sp1 transcription factor shows significantly reduced intensities of {(PPh3)Au} adducts. The results suggest the possibility of systematic (electronic, steric) variations of "carrier" group PR3 and "leaving" group L as well as the nature of the zinc finger in modulation of biological activity. The cytotoxicity, cell cycle signaling effects, and cellular accumulation of the series are also reported. All compounds display cytotoxicity in the micromolar range upon 96 h continuous exposure to human tumor cells. The results may have relevance for the reported inhibition of viral load in simian virus by the gold(I) drug auranofin.

Mesenchymal stem cells (MSC) may act as an alternative for inefficacy of antibiotics and rising antibiotic resistance in endometritis. The present study aimed to cure the chronic endometritic buffaloes using allogenic adipose-derived MSCs (AD-MSC). AD-MSCs were isolated from buffalo adipose tissue and characterized by multilineage differentiation as well as MSC-specific markers. The in vivo safety and efficacy were assessed after infusion of AD-MSCs. In safety trial, cells were administered in healthy buffaloes via different routes (IV and IC) followed by examination of clinical and hematological parameters. In efficacy study, AD-MSCs treatments (IV and IC) and antibiotic therapy (ABT) in endometritic buffaloes were comparatively evaluated. AD-MSCs did not induced any immunological reaction in treated buffaloes. PMN count, CRP levels and VDS were significantly (p ≤ 0.05) reduced after AD-MSCs infusions in IV and IC groups and no significant difference was observed in antibiotic group. The IV group was marked with 50% absolute risk reduction in endometritis and 50% live calf births after artificial insemination in comparison with ABT group. Anti-inflammatory cytokines (IL4 and IL10) and anti-microbial peptides (PI3, CATHL4, LCN2 and CST3) expressions were significantly (p ≤ 0.05) upregulated in IV group. The calf delivery rate after the treatments in IV group was higher (50%, 3 calves) than the other groups (IC: 33.3%, 2 calves; ABT: 16.6%, 1 calf). In conclusion, the administration of AD-MSCs through IV route was found to be safe and efficacious for alleviating chronic endometritis in dairy buffaloes.

Therefore, we aimed to identify local radiographic predictors of Medication-Related Osteonecrosis of the Jaw (MRONJ) in panoramic images of oncologic patients undergoing tooth extraction. Based on a retrospective longitudinal cohort study design, patients were included if undergoing one or more tooth extraction, with at least one administration of ARDs, and presence of pre- and post-operative panoramic radiographs. After data collection, blinded and independent observations were performed. Eleven distinct imaging-related parameters were assessed preoperatively and five postoperatively, at each extraction site. A case-control and subgroup analysis assessing MRONJ development was performed. Significance level is set to 0.05 (5%). A total of 77 oncologic patients were selected, undergoing 218 tooth extractions, from which 63 teeth (29%) in 39 patients (51%) developed MRONJ. Results showed that patients developed significantly more MRONJ with longer ARD treatment (p = 0.057), teeth with absent and incomplete endodontic fillings with caries, widened periodontal ligament space and/or periapical lesions (p = 0.005), and sclerotic and heterogenous bone patterns (p = 0.005). In conclusion, tooth extraction sites presenting with infections and bone sclerosis are at higher risk to develop MRONJ.

While numerical methods like isogeometric analysis (IGA) have been used for modeling the material transport process via solving partial differential equations (PDEs), they require long computation time and huge computation resources to ensure accurate geometry representation and solution, thus limit their biomedical application. Here we present a graph neural network (GNN)-based deep learning model to learn the IGA-based material transport simulation and provide fast material concentration prediction within neurite networks of any topology. Given input boundary conditions and geometry configurations, the well-trained model can predict the dynamical concentration change during the transport process with an average error less than 10% and [Formula: see text] times faster compared to IGA simulations. The effectiveness of the proposed model is demonstrated within several complex neurite networks.

(thyme oils) obtained from Loess Plateau in China had been studied. 130 constituents of thyme oils were determined using gas chromatography-mass spectrometry (GC-MS) and carvacrol ethyl ether was firstly reported as a new natural product, which has been used as a synthetic flavoring substance with no safety concern. The thyme oils showed the anti-oxidant activity using 2,2 diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonate) (ABTS), ferric reducing antioxidant power (FRAP) and thiobarbituric acid reactive substances (TBARS) and conferred protection against oxidative stress in zebrafish. In addition, a class of carvacrol analogues was found to develop as potential natural antioxidant products of thyme oils from Loess Plateau by the correlation analysis. YL-thyme oil performed the best antioxidant activity in this research, which could be recommended as preferred sources of thyme oils. Furthermore, YL-thyme oil exhibited a potent antioxidant capacity by reactive oxygen species (ROS) scavenging, enhancing the endogenous antioxidant system, inhibiting lipid peroxidation and activation of Keap1/Nrf2 pathway in zebrafish.

However, the sequential delivery of multiple proteins acting separately intracellular and extracellular to their sites of action remains a challenge. Here, we construct a nanosystem (PEI-PEG-TRAIL@IONP-GOx) to sequentially release tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) extracellularly and glucose oxidase (GOx) intracellularly for synergistic cancer treatment. The nanosystem is built as a core-shell structure. The core is a pH responsive nanoassembly of boronic acid modified iron oxide nanoparticles (FPBA-IONPs) and polyphenol decorated GOx. The shell is a PEGylated polyethyleneimine (PEI-PEG) polymer on which TRAIL was coupled by a matrix metalloproteinase-2 (MMP-2) responsive peptide. Once the nanosystems were magnetically guided to the tumor site, TRAIL was quickly released by the extracellular MMP-2 to induce tumor apoptosis and enhanced the cellular uptake of the cores. After cytosolic delivery, FPBA-IONPs and GOx were disassembled intracellularly to trigger a cascade reaction to generate free radicals for tumor inhibition. Both in vitro and in vivo experiments proved the separate delivery of TRAIL and GOx and their remarkable synergistic anti-cancer effect. We believe that this nanosystem can offer a new approach for the multistage delivery of proteins and accomplish the objective of protein cooperation for cancer treatment.

Herein, we report the first complete aerobic dehydrogenation pathway to large-scale production of isoquinolines. The discovery of this visible light photoredox reaction was enabled through the combination of mathematical simulations and real-time quantitative mass spectrometry screening. The theoretical calculations showed that hyper-conjugation, the main underlying factor hindering the aerobic oxidation of tetrahydroisoquinolines, is relieved both by π- and σ-donating substituents. This mechanistic insight provided a novel photocatalytic route based on N-substituted auxiliaries that facilitated the conversion of tetrahydroisoquinolines into the corresponding isoquinolines in just three simple steps (yield 71.7% in bulk-solution phase), using unmodified Ru(bpy)3Cl2 photocatalyst, sun energy, atmospheric O2, and at ambient temperature.

The unique properties and potential applications of graphene nanoshells with well-defined small cavities and delicately designed graphene shells are strongly considered. Herein, a mesoscale approach to fabricate graphene nanoshells with a single or few graphene layers and quite small diameters through a catalytic self-limited assembly of nanographene on in situ formed nanoparticles was proposed. The graphene nanoshells with a diameter of ca. 10-30 nm and a pore volume of 1.98 cm(3) g(-1) were employed as hosts to accommodate the sulfur for high-rate lithium-sulfur batteries. A very high initial discharge capacity of 1520 mAh g(-1), corresponding to 91% sulfur utilization rate at 0.1 C, was achieved on a graphene nanoshell/sulfur composite with 62 wt % loading. A very high retention of 70% was maintained when the current density increased from 0.1 C to 2.0 C, and an ultraslow decay rate of 0.06% per cycle during 1000 cycles was detected.

Leg ulceration is one of the complications associated with SCD. There is a gap in the literature in regard to the prevalence of leg ulcers among SCD patients in Saudi Arabia. Objectives The primary objective of this study was to evaluate the lifetime prevalence of leg ulcers in SCD patients in our population and to study the predictive factors of leg ulcers by using sociodemographic factors, clinical manifestations of SCD, and other relevant factors like hydroxyurea. Methods A cross-sectional study design was utilized. Data collection was done using an electronic survey to collect self-reported information for the prevalence of leg ulcers and possible associated factors. The survey was distributed using social media platforms. Chi-square test was used to test for the presence of an association between having leg ulcers and sociodemographic variables as well as SCD related history. Furthermore, binary logistic regression was utilized to determine factors predicting the incidence of leg ulcers among SCD. Results A total of 790 valid responses were included in the study. Among these, 646 were included in the analysis of leg ulcers prevalence. From them, 52 (8%) SCD patients reported a history of leg ulcers. The male to female ratio was (9.7% vs 7.2%). The age group most affected by leg ulcers was those older than 50 (16.7%). There was no significant association between a history of leg ulcers and sociodemographic variables. The only predictive factors for leg ulcers were having six to eight vaso-occlusive crises per month and having more than eight vaso-occlusive crises per month. Conclusion Leg ulcers among SCD patients in Saudi Arabia were considerably prevalent (8%). There was no statistically significant correlation between leg ulceration and sociodemographic variables. Leg ulcers were more likely in patients with a history of highly frequent vaso-occlusive crises. No association was found between the incidence of leg ulcers and other complications of sickle cell disease or hydroxyurea.

Attenuated Total Reflection Fourier Transform Infrared spectroscopy (ATR-FTIR) revealed the existence of higher order assembly of Zn2+: GnRH [6-D-Phe]. Nuclear Magnetic Resonance spectroscopy (NMR) indicated a weak interaction between Zn2+ and GnRH [6-D-Phe]. Atomic Force Microscopy (AFM) showed the existence of GnRH [6-D-Phe] oligomers and fibrils. Molecular Dynamic (MD) simulation of the 10:1 Zn2+: GnRH [6-D-Phe] explored the interaction and dimerization processes. In contrast to already existing short peptide fibrils, GnRH [6-D-Phe] nanostructures and fibrils form in a Tris-buffered pH environment in a controlled manner through a temperature reduction and a pH shift. The lyophilized Zn2+: GnRH [6-D-Phe] assembly was tested as a platform for the sustained delivery of GnRH [6-D-Phe] and incorporated into two different oil vehicle matrices. The in vitro release was slow and continuous over 14 days and not influenced by the oil matrix.

- ArticleLiu S, Tang Q, Huang J, Zhan M, Zhao W, Yang X, Li Y, Qiu L, Zhang F, Lu L, He X.Aging (Albany NY). 2021 04 04;13(8):11257-11280.In order to explore the prognosis of tumor mutation burden (TMB) and the relationship with tumor infiltrating immune cells in hepatocellular carcinoma (HCC), we downloaded somatic mutation data and transcriptome profiles of 376 HCC patients from The Cancer Genome Atlas (TCGA) cohort. We divided the samples into high-TMB and low-TMB groups. A higher TMB level indicated improved overall survival (OS) and was associated with early pathological stages. One hundred and nine differentially expressed genes (DEGs) were identified in HCC. Moreover, based on four hub TMB-related signatures, we constructed a TMB Prognostic model (TMBPM) that possessed good predictive value with area under curve (AUC) of 0.701. HCC patients with higher TMBPM scores showed worse OS outcomes (p < 0.0001). Moreover, DCs subsets not only revealed higher infiltrating abundance in the high-TMB group, but also correlated with worse OS and hazard risk for high-TMB patients in HCC. Meanwhile, CD8+ T cells and B cells were associated with improved survival outcomes. In sum, high TMB indicates good prognosis for HCC and promotes HCC immune infiltration. Hence, DCs and the four hub TMB-related signatures can be used for predicting the prognosis in HCC as supplements to TMB.
