- Advances in chronic lymphocytic leukemia 2013, SpringerChronic lymphocytic leukemia (CLL) is the most common leukemia in the Western world. CLL has a highly varied clinical course. While advances in CLL therapy are noted, many patients still succumb to this illness. Like most progress in medicine, solid advances in the diagnosis, prognosis and treatment of CLL are rooted in an in-depth understanding of the basic and translational biology of CLL. In this book, CLL experts have contributed state-of-the-art summaries of various important aspects of CLL biology and have discussed the translational implication of such findings. This book, which is directed at physicians and researchers alike, aims to educate broadly and deeply. Intentionally, the many aspects and nuances of CLL clinical care that can only really be appreciated through direct patient care are not covered here, but instead, the book presents basic aspects of CLL that underlie many of the contemporary decisions that are made in CLL research and clinical settings. In total, 940 cases of CRS were diagnosed in this population and these patients were subjected to detailed analysis. Hematologic malignancies have recently been identified among the obesity associated malignancies spanning the lifespan from childhood to the elderly and include leukemia, myeloma, lymphoma and others. In addition to the etiologic association between obesity and hematologic malignancies, the presence of obesity has profound effects on therapy by impacting pharmacokinetics of chemotherapeutic agents, dose, adipocyte metabolism and drug distribution. These may be particularly important in hematopoietic stem cell transplantation. Another important aspect of the association of obesity with hematologic malignancies is the increased incidence of obesity in children who successfully complete therapy for acute lymphoblastic leukemia. This and other observations indicate important relations between the hematopoietic systems and fat metabolism. This volume on Energy Balance in Hematologic Malignancies will provide an important volume in this series and a basis for better understanding etiology, mechanisms, therapeutics implications and experimental approaches. This volume of energy balance and cancer will focus on the relation of obesity to hematologic malignancies, the epidemiology, potential mechanisms, and thereapeutic considerations including effects on pharmacologic and physical approaches as well as the delayed effects of therapy on energy balance.
- The growing knowledge about disturbances of epigenetic gene regulation in hematopoietic stem cell disorders such as myeloid leukemias and preleukemias (myelodysplasias) is now being translated into treatment approaches that target the epigenetic defects pharmacologically. Such approaches include reactivation of tumor suppressor genes and other antiproliferative genes by reversal of DNA hypermethylation through azanucleosides, and use of HDAC inhibitors to reverse gene silencing mediated by chimeric, leukemia-specific transcription factors. This book first presents the latest evidence derived from preclinical models regarding the epigenetic regulation of hematopoietic stem cell differentiation and hemoglobin production. The significance of DNA methylation abnormalities in hematopoietic disorders and of epigenetic disturbances in lung cancer and other solid tumors is then discussed. A major part of the book, however, relates specifically to the translation of basic research and drug development to clinical applications, and in this context both present and future clinical strategies are considered. Individual chapters are devoted to the use of DNA hypomethylating agents and chromatin-modifying agents, and the treatment of hematologic malignancies and solid tumors by means of epigenetic agents is discussed in detail.

- Microcirculatory effects of hemoglobin solutions 2004, KargerSolutions of stroma-free hemoglobin have been investigated for their potential as blood replacement fluids for more than 70 years. Despite many attempts to overcome their unwanted side effects through chemical modification of the hemoglobin molecule, none of the potential solutions has been approved for clinical use in Europe or the United States.In recent years, the vasoconstrictive activity of hemoglobin in the plasma was identified as the pivotal problem of hemoglobin-based blood substitutes, compromising nutritional perfusion and thus impeding oxygen unloading at the site of the microcirculation. One of the prevailing assumptions is that the precapillary vasoconstriction and the ensuing tissue underperfusion is caused by the high affinity of free hemoglobin for nitric oxide. To resolve this problem, a number of recombinant techniques involving site-directed mutagenesis as well as several chemical approaches involving polymerization and pegylation have been developed.This volume summarizes the latest research on the effects of some of these new hemoglobin solutions on the microvasculature and tissue oxygenation. It is recommended reading for all those interested in finding alternatives for donor blood in transfusion medicine, including emergency specialists, anesthesiologists, surgeons, trauma surgeons and other clinicians who are frequently confronted with blood loss and the need for blood replacement
