Brison, editors.The book considers signaling events from the zygote embryo through to the blastocyst with relevant data from embryonic stem (ES) cells, including dialogue with the extracellular environment and with the maternal tract during the implantation process. Application of the knowledge described to improve the success of human and animal assisted conception is considered where appropriate, but the focus is largely on fundamental rather than applied cell/molecular biology, as this is the area that has historically been neglected. While the general features of metabolism during preimplantation development are well established, especially in terms of nutrient requirements, uptake and fate, remarkably little is known about early embryo signaling events, intracellular or intercellular, between individual embryos in vitro or with the female reproductive tract in vivo. This contrasts with the wealth of information on cell signaling in somatic cells and tissues, as a glance at any textbook of biochemistry illustrates. This lack of information is such that our understanding of the molecular cell biology of early embryos -- a prerequisite to defining the mechanisms which regulate development at this critical stage of the life cycle -- is seriously incomplete. This volume is the first to address this issue by describing the current state of knowledge on cell signaling during mammalian early embryo development and highlighting priority areas for research.Also available: Print – 2015

- 2013Chien-Jung Lin.During embryonic development, dynamic remodeling of the chromatin landscape is required for the transcriptional machinery to access the genomic DNA in a spatiotemporally controlled manner. This intricate level of regulation allows precise gene regulation, necessary for complex morphogenetic processes. The Brg1/Brm-associated factor (BAF) complex is a multisubunit, ATP-dependent chromatin remodeling complex. Among the subunits, Brg1 is an ATPase that is critical for the function of the complex. Here, we study the role of Brg1 in foregut and cardiac outflow tract development. In the foregut, Brg1 functions with HDAC proteins in the ventral epithelium to regulate Nkx2-1 expression, essential for foregut septation into trachea and esophagus. In the absence of Brg1, embryos develop a single-tube foregut, reminiscent of human patients with tracheoesophageal fistula. In the developing heart, Brg1 functions in the secondary heart field-derived myocardium to control Semaphorin 3c (Sema3c) expression, essential for the outflow tract to septate into pulmonary artery and aorta. Deletion of Brg1 in the secondary heart field results in decreased neural crest cell colonization of the outflow tract, leading to a phenotype similar to the persistent truncus arteriosus pathology. Brg1 cooperates with HDAC proteins and Chd7 in the outflow tract myocardium to control Sema3c expression. These studies demonstrate novel functions and molecular mechanisms of Brg1 in foregut and cardiovascular development.

- Investigating Wnt/ss-catenin signaling in stem cells and progenitors of the developing and adult mouse brain2012Angela N. Bowman.In order for a tissue to be properly sized and patterned, enormous care must be taken to ensure that cell proliferation and differentiation are tightly regulated. The embryonic brain must execute a temporally and spatially defined developmental scheme, resulting in the production of millions of cells of multiple lineages. These cells and their resultant functional architecture must be maintained in the homeostatic adult mouse brain. These processes are in part controlled by external signaling factors, which include Wnts, and their transcriptional programs. However, the degree to which and exact mechanisms by which cell fate choices--including self-renewal, proliferation, and differentiation--are controlled by Wnt/ss-catenin signaling are unknown. Here, I describe studies to investigate these questions. First, I show the identification of a novel target of Wnt/sscatenin signaling, Pou6f1, which mediates neural differentiation and proliferation in embryonic neural progenitor cells. Next, I illustrate the use of a lineage tracing approach using a Wnt/ss-catenin target gene, Axin2, to identify and trace sites of Wnt/ss-catenin responsiveness in the developing and adult mouse brain and central nervous system. Using this same Axin2CreERT2 mouse, I describe the functional consequences of loss of sscatenin from Axin2-expressing cells. Axin2CreERT2 labels numerous radial glial cell populations throughout the central nervous system, three of which serve as functional stem/progenitor populations. Loss of ss-catenin from adult Axin2+ populations results in tissue architecture defects and increased proliferation, presumably because Wnt serves as a self-renewal signal. These studies demonstrate the importance of Wnt/ss-catenin signaling for mediating multiple cell fate choices during the development and homeostasis of the mouse central nervous system, and suggest mechanisms by which neural stem/progenitor populations could be manipulated for therapeutic outcomes.

Oocyte and embryo donation has established itself as a mainstay procedure within assisted reproductive care, and the breadth, depth and complexity of practice is deserving of focused attention. Much has changed within the field of oocyte and embryo donation since the publication of the first edition of Principles of Oocyte and Embryo Donation in 1998, thus the need for a completely updated and more expansive text. The second edition of this book provides an overview of the major issues affecting men and women engaged in the practice of oocyte and embryo donation. A primary emphasis has been placed on defining the standards of practice that have evolved over the past 30 years, clearly stating the outcomes expected from adhering to these established protocols. Details of both the basic science and the clinical medicine are presented together and attention is also focused on the non-reproductive aspects inherent to this unique method of assisted reproduction that involves opinions from lawyers, ethicists, mental health care professionals and theologians. Oocyte and embryo donation requires a working knowledge of the medicine, the law and the ethics that underlies its foundation. - Regulation of adult stem cell function and identity by a histone variant and polycomb group proteins in the drosophila testis2012Jose Rafael Morillo Prado.Many adult tissues with short-lived cells such as blood and skin require a constant replacement of aging cells. This process is often accomplished by adult stem cells, both self-renew and give rise to differentiating progeny. The process of self-renewal and differentiation must be tightly controlled: failure to self-renew results in the loss of the stem cell population, and lack of differentiation impairs tissue function with dire consequences to the organism. The fate decisions of stem cells are guided by the interaction between extracellular signals from the local microenvironment and intrinsic factors that set the state of the cell, many of which are coordinated by transcriptional programs that are, in turn, is controlled by posttranslational modifications of histones or replacement of canonical histones with histone variants. Polycomb group (PcG) proteins partake in the modification of histones and influence the transcriptional state of genes. PcG proteins are involved in transcriptional regulation by inducing the methylation and ubiquitination of histone tails, which maintain the repression of target genes through cell divisions. Emerging evidence indicates that replacement of canonical histones with histone variants is also important in cell fate decisions. Here I show that PcG proteins Psc and Su(z)2 restrict proliferation and maintain the identity of the cyst stem cell (CySC) lineage in the Drosophila testis. Loss of Psc and Su(z)2 gene function in CySC but not in the germline stem cell (GSC) lineage resulted in the formation of a tumor composed of proliferative mutant cells of abnormal identity, likely due to the derepression of the Hox gene Abdominal-B. The resulting formation of tumor-like masses of proliferative cells interfered non-cell autonomously with maintenance of the germline most likely by displacing GSCs from their niche. I also show that the histone variant His2Av is required cell-autonomously for maintenance of adult stem cells in both somatic and germ cell lineages. GSCs lacking His2Av function were lost over time. Surprisingly, GSCs null mutant for His2Av gave rise to germ cells that underwent normal transit amplification divisions and differentiation to spermatocytes and spermatids. Similarly, CySCs lacking His2Av were also lost over time, but were able to give rise to differentiated cyst cells. My results indicate that His2Av and Stat92E likely maintain GSCs and CySCs independently. His2Av mutant GSCs expressed Stat92E and had properly oriented centrosomes, while Stat92E mutant GSCs expressed His2Av. My results expand knowledge chromatin states required for adult stem cell function, showing, for the first time, that a histone variant has a unique role in maintaining stem cells.

- 2012Xiaohu Ouyang.Embryonic development and tissue regeneration are regulated by complex genetic programs that are dynamically executed in a stereotypic manner, and deciphering these molecular mechanisms requires the ability to control gene function with spatial and temporal precision. Synthetic chemistry has been increasingly used to create reagents that can enable such genetic manipulations. In this thesis I describe the development and application of caged morpholino (cMO) oligonucleotides and small molecules to study zebrafish embryonic development and larvae tail regeneration. In Chapter 1, we survey how chemical tools and engineered proteins have been used to perturb developmental processes at the DNA, RNA, protein, and cellular levels. We discuss the design principles, experimental capabilities, and limitations of each method, as well as future challenges for the chemical and developmental biology communities. In Chapter 2, we describe optimized methods for the design and synthesis of hairpin cMO oligonucleotides incorporating a dimethoxynitrobenzyl (DMNB)-based bifunctional linker that permits cMO assembly in only three steps from commercially available reagents. Using this simplified procedure, we have systematically prepared cMOs with differing structural configurations and investigated how the in vitro thermodynamic properties of these reagents correlate with their in vivo activities. Through these studies, we have established general principles for cMO design and successfully applied them to several developmental genes. Our optimized synthetic and design methodologies have also enabled us to prepare a next-generation cMO that contains a bromohydroxyquinoline (BHQ)-based linker for two-photon uncaging. Collectively, these advances establish the generality of cMO technologies and will facilitate the application of these chemical probes in vivo for functional genomic studies. In Chapter 3, I first describe our genome-wide microarray analysis and whole-mount in situ hybridization studies that led us to the identification of 18 genes, including hyaluronan synthase 3 (has3), that were expressed during zebrafish larvae tail regeneration. I also report the functional studies of hyaluronan (HA) synthesis using small molecule inhibitor 4-methylumbelliferone (4-MU), in which we showed that 4-MU inhibited tail regeneration by preventing injury-induced cell proliferation. In addition, 4-MU also reduced the expressions of dlx5a and junbb, markers of the wound epithelium and blastema-like mesenchymal cells. We also illustrated that simultaneous dosing of Gsk3[beta] inhibitor BIO and 4-MU could rescue the 4-MU-induced regeneration defects. Our results indicated that the crosstalk between hyaluronan synthesis and Gsk3[beta] signaling played an important role in regulating tail regeneration.
