    G protein-coupled receptors (GPCRs) are heptahelical transmembrane receptors that convert extra-cellular stimuli into intra-cellular signaling, and ultimately into biological responses. Since GPCRs are natural targets for approximately 40% of all modern medicines, it is not surprising that they have been the subject of intense research. Notwithstanding the amount of data generated over the years, discovering ligands of these receptors with optimal therapeutic properties is not straightforward and has certainly been hampered for years by the lack of high-resolution structural information about these receptors.--Back cover.

    Emerging evidence indicates that the Gadd45 family of genes play a unique and critical role as sensors of stress, including genotoxic, physiological and oncogenic stress. The stress response Gadd45 family of genes (Gadd45a, Gadd45b & Gadd45g), discovered by Dr. Liebermann and other researchers, encode for small (18 kd) nuclear/cytoplasmic proteins).These genes are rapidly induced by a wide variety of endogenous and exogenous stress stimuli. In spite of marked similarities, Gadd45 genes are regulated differently & exhibit functional diversity. Gadd45 are implicated in cell cycle arrest, DNA demethylation & repair, apoptosis, cell survival, genomic stability, inflammation, & in response to physiological and oncogenic stress. Functions of Gadd45 genes are mediated by protein-protein interactions that modulate structure/function of other cellular proteins implicated in cell cycle regulation and the response of cells to stress; these interactions vary depending upon the biological setting including cell type, developmental stage and stress/stimulus.     Sickle Cell Anemia, Beta Thalassemia, Severe Combined Immunodeficiency (SCID), and Type I Diabetes share one commonality: these diverse disorders can all be attributed to faulty hematolymphoid effector cells which are largely caused by genetic mutations that alter hematopoietic cell-intrinsic function. These defective blood cells inherit their genetic deficiencies from hematopoietic stem cells (HSC) as they differentiate. Thus, each of these unique diseases should be theoretically curable through the same strategy: replacement of patients' HSCs carrying the problematic mutation with normal HSCs from disease-free donors, thereby generating entire new, healthy hematolymphoid systems. Replacement of disease-causing stem cells with healthy ones has been achieved clinically via hematopoietic cell transplantation (HCT) for the last 50 years, as a treatment modality for a variety of cancers and immunodeficiencies with moderate, but increasing success. However this technique bears high morbidity and mortality severely limiting its utilization. Purified hematopoietic stem cell transplantation is associated with high toxicity primarily attributed to the myeloablative pre-conditioning necessary to facilitate engraftment of the donor HSCs. However the function and mechanistic role of these reagents has been up until now poorly understood. This thesis deconstructs the current barriers in HCT and pure HSC transplantation, and suggests that in addition to the immune barrier, hematopoietic stem cell niche availability is another critical obstacle that needs to be overcome to allow for efficient hematolymphoid replacement. In this thesis I show that in the homeostatic state, most hematopoietic stem cell niches are occupied by host HSCs limiting the ability of incoming HSC engraftment. However, a limited number of HSC niches are vacated by HSC that are egressing from the peripheral blood into the bone marrow in a division independent manner, allowing for true but minimal HSC engraftment without myeloablative pre-conditioning. This thesis illustrates new, up-and-coming strategies to combat the obstacle of HSC niche availability. Here I prove that one non-myeloablative conditioning approach that capitalizes upon physiological HSC egress is repetitive transplantation of purified HSCs. However, this approach is rather laborious and inefficient, and I show that another more efficient novel non-myeloablative conditioning approach to vacate the HSC niche is antibody-mediated depletion of host HSCs. Furthermore, in this thesis I demonstrate that antibody-mediated inhibition of c-kit effectively depletes host HSCs and allows for efficient, non-toxic HSC transplantation. These novel non-myeloablative conditioning methods illustrate new means of overcoming the current barriers to hematolymphoid replacement thereby allowing for the potential expansion of this technique to treat a variety of inherited diseases of hematopoietic and immune function.

    The interest in using legged robots for a variety of terrestrial and space applications has grown steadily since the 1960s. At the present time, a large fraction of these robots relies on electric motors at the joints to achieve mobility. The load distributions inherent to walking, coupled with design constraints, can cause the motors to operate near their maximum torque capabilities or even reach saturation. This is especially true in applications like space exploration, where critical mass and power constraints limit the size of the actuators. Consequently, these robots can benefit greatly from motion optimization algorithms that guarantee successful walking with maximum margin to saturation. Previous gait optimization techniques have emphasized minimization of power requirements, but have not addressed the problem of saturation directly. This dissertation describes gait optimization techniques specifically designed to enable operation as far as possible from saturation during walking. The benefits include increasing the payload mass, preserving actuation capabilities to react to unforeseen events, preventing damage to hardware due to excessive loading, and reducing the size of the motors. The techniques developed in this work follow the approach of optimizing a reference gait one move at a time. As a result, they are applicable to a large variety of purpose-specific gaits, as well as to the more general problem of single pose optimization for multi-limbed walking and climbing robots. The first part of this work explores a zero-interaction technique that was formulated to increase the margin to saturation through optimal displacements of the robot's body in 3D space. Zero-interaction occurs when the robot applies forces only to sustain its weight, without squeezing the ground. The optimization presented here produces a swaying motion of the body while preserving the original footfall locations. Optimal displacements are found by solving a nonlinear optimization problem using sequential quadratic programming (SQP). Improvements of over 20% in the margin to saturation throughout the gait were achieved with this approach in simulation and experiments. The zero-interaction technique is the safest in the absence of precise knowledge of the contact mechanical properties and friction coefficients. The second part of the dissertation presents a technique that uses the null space of contact forces to achieve greater saturation margins. Interaction forces can significantly contribute to saturation prevention by redirecting the net contact force relative to critical joints. A method to obtain the optimal distribution of forces for a given pose via linear programming (LP) is presented. This can be applied directly to the reference gait, or combined with swaying motion. Improvements of up to 60% were observed in simulation by combining the null space with sway. The zero-interaction technique was implemented and validated on the All Terrain Hex-Limbed Extra-Terrestrial Explorer (ATHLETE), a hexapod robot developed by NASA for the transport of heavy cargo on the surface of the moon. Experiments with ATHLETE were conducted at the Jet Propulsion Laboratory in Pasadena, California, confirming the benefits predicted in simulation. The results of these experiments are also presented and discussed in this dissertation.

    Gamma-Glutamyl Transpeptidases (g-GTs) are members of the N-terminal nucleophile hydrolase superfamily, enzymes that cleave the g-glutamyl amide bond of glutathione to liberate cysteinylglycine. The released g-glutamyl group can be transferred to water (hydrolysis) or to amino acids or short peptides (transpeptidation). g-GT plays a key role in the gamma glutamyl cycle by regulating the cellular levels of the antioxidant glutathione, hence it is a critical enzyme in maintaining cellular redox homeostasis.g-GT is upregulated during inflammation and in several human tumors, and it is involved in many physiological disorders related to oxidative stress, such as Parkinson's disease and diabetes. Furthermore, this enzyme is used as a marker of liver disease and cancer. This book covers current knowledge about the structure-function relationship of g-GTs and gives information about applications of g-GTs in different fields ranging from clinical biochemistry to biotechnology and biomedicine.

    Stress is a fundamental aspect of aging as accumulated damage from a lifetime of stress can limit lifespan, and protective responses to stress can extend lifespan. In this study, we identify a conserved C. elegans GATA transcription factor, egl-27, that is involved in several stress responses and aging. We found that overexpression of egl-27 extends the lifespan of wild-type animals. Furthermore, egl-27 is required for the pro-longevity effects from impaired insulin/IGF-1 like signaling (IIS), as reduced egl-27 activity fully suppresses the longevity of worms that are mutant for the IIS receptor, daf-2. egl-27 expression is inhibited by daf-2 and activated by pro-longevity factors daf-16/FOXO and elt-3/GATA, suggesting that egl-27 acts at the intersection of IIS and GATA pathways to extend lifespan. Consistent with its role in IIS signaling, we found that egl-27 is involved in stress response pathways. egl-27 expression is induced in the presence of multiple stresses, its targets are significantly enriched for many types of stress genes, and altering levels of egl-27 itself affects survival to heat and oxidative stress. Finally, we found that egl-27 expression increases between young and old animals, suggesting that increased levels of egl-27 in aged animals may act to promote stress resistance. These results identify egl-27 as a novel factor that links stress and aging pathways.

    China's concentrated HIV epidemic is on the brink of becoming a generalized one and syphilis infection has become a major public health threat. Social factors relating to gender and gender inequality exacerbate the spread of HIV and sexually transmitted infections (STI) in China. A better understanding of the proximate social determinants of HIV related to gender will be crucial to effectively curbing HIV and other STIs in China. Aspects of China's governance - including administrative procedures, the developing legal system, social institutions, and the public health infrastructure are instrumental in shaping strategies and responses to HIV. International studies suggest that women who are more economically and socially vulnerable may also have a greater risk of HIV infection, yet few initiatives have focused on discrete areas where achievable and sustainable gender policy measures could be linked to the public health response. This study presents perspectives ranging from criminology to social psychology to better understand how gender perspectives can inform HIV policy in the context of China. - Publisher's Description.

    The physiological state of the genome is chromatin. Chromatin is a dynamic polymer composed of genomic DNA, histone proteins, and other factors. Dysregulation of chromatin homeostasis can lead to diverse human pathologies including cancer. This work focuses on chromatin-based mechanisms of gene regulation; more specifically the (1) targeting and (2) stabilization of gene-regulatory proteins on the chromatin template. In regard to chromatin targeting, not much is known about how gene-regulatory proteins selectively associate with their targets. Herein we detail the discovery of a new targeting factor, the nuclear phospholipid phosphatidylinositol-5-phosphate (PtdIns(5)P). PtdIns(5)P directly interacts with the tumor suppressor ING2 (inhibitor of growth family member 2) in the nucleus to coordinate gene expression of select ING2 targets. In regard to chromatin stabilization, lysine methylation is a principle mechanism for retaining chromatin-effector modules at discrete chromatin zones. Numerous lysine methylation events have been discovered on the major protein component of chromatin, histones, but relatively few specific modules have been characterized to sense these modifications. We screened through a library of putative methyl-lysine binding domains, which resulted in the identification of three novel chromatin binding modules. Taken together these data shed insight into the molecular modes of action of gene regulation by chromatin and phosphoinositide pathways.

    The 1st World Congress on Geriatrics and Neurodegenerative Disease Research (GeNeDis 2014), will focus on recent advances in geriatrics and neurodegeneration, ranging from basic science to clinical and pharmaceutical developments and will provide an international forum for the latest scientific discoveries, medical practices and care initiatives. Advances information technologies will be discussed along with their implications for various research, implementation and policy concerns. In addition, the conference will address European and global issues in the funding of long-term care and medico-social policies regarding elderly people. GeNeDis 2014 takes place in Corfu, Greece, 10-13 April 2014. This volume focuses on the sessions that address geriatrics.

    All European countries, including those that have recently joined the European Union or are candidates for membership, currently show a clear trend towards an increasing number of accidental injuries. This holds true for a range of injuries, including accidents among the elderly, sports injuries, and trauma due to traffic accidents. The increase in the number of injuries is accompanied by rising expectations among patients, who anticipate good functional results even after serious injuries. Despite these developments, trauma surgery is not yet established as an independent field in all European countries. Against this background, there is a clear need for a book that covers the state of the art in trauma surgery. This volume, which focuses on basics in trauma surgery is intended to help to meet this need. It will also serve to harmonize the practice of trauma surgery within the European Union and to prepare for the UEMS EBSQ trauma surgery.

    Generic drugs are now familiar objects in clinics, drugstores, and households around the world. We like to think of these tablets, capsules, patches, and ointments as interchangeable with their brand-name counterparts: why pay more for the same? And yet they are not quite the same. They differ in price, in place of origin, in color, shape, and size, in the dyes, binders, fillers, and coatings used, and in a host of other ways. Claims of generic equivalence, as physician-historian Jeremy Greene reveals, are never based on being identical to the original drug in all respects, but in being the same in all ways that matter. How do we know what parts of a pill really matter? Decisions about which differences are significant and which are trivial in the world of therapeutics are not resolved by simple chemical or biological assays alone. Questions of therapeutic similarity and difference are also always questions of pharmacology and physiology, of economics and politics, of morality and belief. Greene chronicles the social, political, and cultural history of generic drugs, narrating the evolution of the generic drug industry from a set of mid-twentieth-century "schlock houses" and "counterfeiters" into an agile and surprisingly powerful set of multinational corporations in the early twenty-first century. The substitution of bioequivalent generic drugs for more expensive brand-name products is a rare success story in a field of failed attempts to deliver equivalent value in health care for a lower price. Greene's history sheds light on the controversies shadowing the success of generics: problems with the generalizability of medical knowledge, the fragile role of science in public policy, and the increasing role of industry, marketing, and consumer logics in late-twentieth-century and early twenty-first century health care.
